实验材料:大豆种子表面消毒后萌发,抗病材料Williams82和感病材料Harosoy生长2周后接种大 豆疫霉菌,接种8小时后取根部。共四个样品(Harosoy mock, Harosoy infected,Williams 82 mock and Williams 82 infected)。
测序方法: HiSeq2000,SE50bp;
数据量: 每个样品10-13M
结果分析:利用STTM技术(短串联靶向模拟),靶向降解内源的miR393发现,抗病材料Williams82中靶向降解miR393后,变为感病。
接种大豆疫霉菌后phasiRNA的分析结果表明侵染后phasiRNA调控NB-LRR和PPR基因的表达。
